Overview

DSIP (Delta Sleep-Inducing Peptide) is a naturally occurring nonapeptide first isolated from rabbit brain tissue during slow-wave sleep in 1977 by Schoenenberger and Monnier. It is found in both free and bound forms in the hypothalamus, pituitary, and various peripheral organs. DSIP is one of the most studied neuropeptides in sleep research.

Research Summary

DSIP has been extensively studied in preclinical sleep research models. Studies in rodent and rabbit models have examined its effects on sleep architecture, particularly delta wave (slow-wave) sleep induction and consolidation. Research has also investigated its interaction with the hypothalamic-pituitary-adrenal (HPA) axis, stress response modulation, and its effects on cortisol and ACTH rhythms. In vitro studies have examined its opioid-modulatory properties and effects on neurotransmitter metabolism. Limited human studies were conducted in the 1980s and 1990s but have not met modern clinical trial standards.

Proposed Mechanism of Action

DSIP is proposed to modulate sleep through multiple mechanisms including interaction with GABAergic systems, modulation of serotonin and dopamine metabolism, and influence on the circadian pacemaker in the suprachiasmatic nucleus. Research also indicates potential interaction with opioid receptor systems and modulation of the HPA axis stress response.
