Overview

Semax is a synthetic heptapeptide derived from a fragment of adrenocorticotropic hormone (ACTH 4-10) with an added Pro-Gly-Pro C-terminal sequence for enhanced metabolic stability. Developed at the Institute of Molecular Genetics of the Russian Academy of Sciences alongside Selank, Semax has been investigated in preclinical research for its neurotrophic and neuroprotective properties.

Research Summary

Preclinical studies on Semax have focused on its neurotrophic properties, including upregulation of BDNF and TrkB receptor expression in rodent brain tissue. Research has examined its effects on neuronal survival in ischemic models, gene expression changes in the hippocampus and prefrontal cortex, and modulation of dopaminergic and serotonergic systems. In vitro studies have documented its effects on neurite outgrowth and neuronal differentiation. Semax is registered as a pharmaceutical agent in Russia for specific indications but has not been evaluated by the FDA.

Proposed Mechanism of Action

Semax is proposed to act through melanocortin receptor subtypes (MC4R, MC3R) while also independently modulating neurotrophic factor expression. Research indicates it upregulates BDNF/TrkB signaling, influences cholinergic and dopaminergic neurotransmission, and may modulate gene expression patterns involved in synaptic plasticity and neuronal survival.
